Who knows about the Rule of Four and what the gist of it is? I flicked through the novel in a bookstore and immediately knew that I wasn't going to read it (not my style), but I am curious enough to want a condensed synopsis of what it is all about.

ya got me. im still trying to figure out why people believe in the "rule of three", "power of three", or "three fold return" or whatever. it seems to hold no water or valid reference to reality for me.

The novel charts the relationship between the four roommates and how obsession can be both a boon and a burden. It is a story about growing up as much as solving the mystery of the Hypnerotomachia Poliphili. The disciplines of Renaissance science, history, architecture, and art are drawn upon to solve the mystery.
It may refer to the four years of a traditional college, or to the four disciplines. It may refer to the four roommates as well.

When I was growing up the old timers(like I am now), said that bad luck comes in three's & it does most of the time. But there be a reason , there are three cycles of the universe, three is the main cycle of the universe , the other two are seven & eleven, everything in the universe opperates on one or more of these cycles. Read the Druids three's. But I have no idea what the rule of four is?

how about the law of fives? add a number save a world. take one away and get hurled (across time and space of course).

I have never ever heard of a "Rule of Four". What book were you looking at? I'm assuming it's fiction because you said it was a novel. Therefore, I'd take it with a grain of salt.
